Câu chuyện bắt đầu ở Nhật thời Chiến Quốc (Sengoku Jidai). Các lãnh chúa giàu có tranh hùng xưng bá, gây nên chiến tranh liên miên không ngừng. Lại thêm nạn yêu quái hòanh hành, khiến người dân nghèo đói khổ cực.

InuYasha – khuyển dạ xoa – tấn công một ngôi làng nhỏ cướp ngọc Tứ Hồn (Shikon no Tama - The Four-Soul Jewel), bị pháp sư trưởng làng Kikyo bắn mũi tên phong ấn vào ngực. Để bảo vệ ngọc Tứ Hồn luôn trong sạch, Kikyo (lúc này đã bị thương nặng) yêu cầu dân làng hỏa thiêu cô cùng viên ngọc…
 
550 năm trôi qua, ở nước Nhật hiện đại, câu chuyện được tiếp tục trong ngày sinh nhật lần thứ 15 của Kagome Higurashi – nữ sinh trung học – sống hạnh phúc cùng gia đình trong căn nhà nằm trong khuôn viên của ngôi đền cổ. Quà sinh nhật ông nội dành cho cô bé chính là viên ngọc Tứ Hồn, và vô tình cô bé đi xuyên qua cái giếng cạn trong đền thờ, trở về thời chiến quốc, tại ngôi làng có InuYasha bị phong ấn. 
Mọi việc thêm rắc rối khi người dân làng đều cho rằng Kagome là kiếp sau của Kikyou.
 Đúng lúc này, ngôi làng bị yêu quái tấn công. Kagome bỏ chạy mang theo viên ngọc. Tình cờ cô chạy lạc vào rừng nơi Inu-Yasha bị phong ấn suốt 50 năm. Kagome rút mũi tên, đưa Inu-Yasha quay lại với cuộc sống. Từ lúc đó, Kagome và Inu-Yasha bắt đầu nhiệm vụ quan trọng: tìm kiếm những mảnh ngọc Tứ Hồn đã bị thất lạc sau khi viên ngọc bị vỡ, bảo vệ và thanh tẩy chúng, không cho chúng rơi vào tay các yêu quái khác.
